Abstract

Conjugative bacteriocin plasmid pPD1 (59kb) of Enterococcus faecalis has been mapped physically by the relational clone method. pPD1 consisted of ten EcoRI fragments with corresponding molecular sizes of 22.5, 13.0, 9.0, 6.9, 5.0, 1.0, 0.6, 0.36, 0.28, and 0.23kb, and in the order of A G C E B F D J H I in the clockwise orientation. Transposition of Tn917 (Em) to pPD1 gave rise to mutants altered in bacteriocin expression. Transposon mutagenesis and the expression of bacteriocin of the relational clones revealed the region of the pPD1 map required for bacteriocin synthesis. Tn917 insertions within a certain region resulted in constitutive clumping, and the DNA sequence was analyzed in the region. The region of pPD1 spans about 8kb and was found to contain ten open reading frames (ORF). Significant homologies of sequence were identified between seven ORFs and genes in the regulatory region of pAD1 and pCF10, respectively.
