Abstract

Analysis of enzyme inhibition and receptor-binding data shows that the average strengths of ionic, hydrogen and van der Waals bonds in aqueous solution are of the order 10, 3 and 1 kcal mol −1 respectively. Peter Andrews illustrates the relevance of these numbers to drug design with back-of-the-envelope calculations on data from a selection of recent papers.
