Abstract

1. The well-known disadvantages in the use of freeze-dried-paraffin-embedded tissues, such as loss and displacements of tissue elements as well as difficulties in flattening sections can be prevented by embedding in glycol methacrylate (GMA). 2. Usual histological stainings and the histochemical demonstration of RNA, DNA and glycogen can be performed without removing the plastic material. The activity of acid and alkaline phosphatase, ATPase and 5-nucleotidase can be demonstrated without difficulty. 3. Lysosomal ali-esterases can be demonstrated with the indoxyl acetate method, while the E-600-sensitive microsomal esterases do react extremely weak. The reactivity of succinodehydrogenase is abolished. Immunohistological reactions to reveal human IgG are strongly diminished.
