Abstract
A number of organic peroxides and hydroperoxides mediate tumor promotion and malignant conversion in mouse skin and are useful probes for defining the role of free radicals in multistage carcinogenesis. Benzoyl peroxide undergoes copper-dependent activation to benzoyloxyl and alkyl radicals in keratinocytes. The benzoyloxyl radical is thought to mediate DNA damage by benzoyl peroxide. Mutations induced by benzoyl peroxide occur at guanine and cytidine bases, particularly in polyguanosines, and may result from site-specific binding of copper to DNA.
