Abstract

Curcumin and β-carotene are two kinds of pigments serve as antitumor, anticancer and antioxidant agents. Theantioxidant activity of curcumin is stronger and more stable as compared to β-carotene, so it can protect β-carotene degradation from light and oxygen. Therefore curcumin has higher antioxidant activity as well. Theresearch was conducted by mixing curcumin and β-carotene in several molarity ratios followed by iradiation usingvolpi lamp for 0, 10, 30, 60, and 90 minutes. The result shows that curcumin has photoprotection effect because itcan protect photooxidation of β-carotene. Mixing of curcumin and β-carotene 1:1 was the most stable ratio forprotecting β-carotene.

BAHAN DAN METODE

Bahan-bahan kimia yang digunakan adalah produk dari Merck diantaranya aseton, heksana, dietil eter, petroleum eter, metanol, etanol, kloroform, Na2SO4 anhidrat, CaCO3, silika gel 60 F254 dan larutan DPPH, serta pigmen standar. Spirulina diekstraksi menggunakan campuran pelarut aseton dan metanol (3:7 v/v) dengan nisbah sampel terhadap pelarut 1:10 (w/v). 3 kemudian filtrat dipartisi menggunakan dietil eter dengan nisbah sampel terhadap pelarut 1:1 (v/v). Β-Karoten dan kurkumin masingmasing dimurnikan dengan kromatografi kolom menggunakan fase diam dan fase gerak yang sesuai. Fase diam yang digunakan adalah silika gel 60 F dan fase gerak kloroform 100% 254 serta kloroform:metanol (95:5 v/v). Fase diam untuk isolasi β-karoten adalah silika gel 60 F dan fase gerak 254 aseton:eter:heksana (2:3:6 v/v). Pigmen β-karoten dan kurkumin masing-masing dilarutkan dalam aseton.
