Abstract

We are developing a toolkit of fluorescent probes for quantifying the protein regulation pathways of living cells at high resolution. This biosensor development program blends genetics, protein structure, nucleic acid structure and fluorescent dye chemistry. Our initial biosensor platform consists of two well characterized single chain antibody classes that bind fluorogen dyes with high affinity and large fluorescence increases. One antibody binds the green‐fluorescing TO1 fluorogen and the other binds Malachite Green to give red fluorescence. We are using this platform to create sensors for protein‐protein interactions, protease activity and membrane surface labeling. Imaging and informatics technologies are integrated with the new fluorescent probe technologies so that these combined tools to obtain spatial and temporal information about pathways in living cells.
