Abstract

Plasmodiophora brassicae Woron is most devastating pathogen causing clubroot of cruciferous crops. The present study used fungicide fluazinam to treat clubroot in order to understand microbial communities. The microbial communities in clubroot affected cabbage rhizosphere with fluazinam was not studied before. Biolog™ MicroPlates and 454 pyrosequencing was implied to study effect of fluazinam on soil microbial community in cabbage rhizosphere to survey clubroot incidence. The disease index (DI) was reduced to 0.95% with fungicide treatment, and the control efficiency was recorded as 98.72%. Dominant taxonomic phyla found among bacteria was Acidobacteria, Actinobacteria, Bacteroidetes, Firmicutes, Gemmatimonadetes, and Proteobacteria. Among fungi, Ascomycota, Basidiomycota and early diverging fungal lineages were reported. Microbial communities in soil reduces greatly at start of two growth stages, but they recovered slowly at rosette stage. We come to conclusion that in Chinese cabbage rhizosphere, the fluazinam effect on fungal community was just transient. High-throughput 454 pyrosequencing could be suitable method of microbial communities’ characterization of cabbage rhizosphere. Moreover, the different microbial communities observed may have potential to improve the clubroot disease management strategies in future.
