Abstract

A flow-injection system with a separation column and an immobilized enzyme reactor is described for the determination of 1,5-anhydro- d-glucitol in serum. Serum samples diluted with water are injected into an anion-exchange column (4 cm × 4 mm i.d.). The eluate is passed through an immobilized pyranose oxidase reactor (5 cm × 4 mm i.d.). the hydrogen peroxide produced is detected chemiluminometrically with a luminol-hexacyanoferrate(III) reaction. The calibration graph is linear over the range 4 × 10 −7−2 × 10 −4 M; the detection limit is 2 × 10 −7 M. The sample throughout is 20 h −1 with a relative standard deviation of less than 2.5%.
