Abstract

Maize is an important cereal crop and occupies an area of 9.25 million ha with production of 24.17 Mt in India. Incidence of 1015% of leaf redness symptoms was observed in maize plants at Siot, Rajouri district of Jammu and Kashmir state in September, 2016. Total DNAs extracted from symptomatic and symptomless maize plants were analysed by nested PCR assay using P1/P7 followed by R16F2n/R16R2 primers. Amplicons of about 1.2 kb were obtained in samples of symptomatic maize plants but not from those of symptomless ones. The 16S rRNA gene sequences of maize leaf redness phytoplasma (MLRP) strains shared 100% nucleotide sequence identity with each other and clustered with phytoplasma strains of 16SrI-B group in phylogenetic analysis. Virtual RFLP analysis, carried out by iPhyClassifier, showed that RFLP profile of MLRP was consistent with that of strains of 16SrI-B subgroup. Based on the 16S rDNA sequence similarity and virtual RFLP pattern, the phytoplasma strains associated with MLRP were identified as ‘Candidatus Phytoplasma asteris’-related. To the best of knowledge, this is the first report of association of ‘Ca. P. asteris’ with a leaf redness disease of maize in India.
