Abstract

Suspensor cells in the embryo of Phaseolus coccineus contain giant chromosomes. The relative DNA content of individual suspensor nuclei was determined by microspectrophotometry of Feulgen-stained squashes. Nuclei of the embryo proper contained only two levels of DNA, 2C and 4C. Cells at the embryo-suspensor junction contained nuclei with amounts of DNA that fell into a regular series 2C, 4C, 8C, 16C, 32C and 64C. In the largest basal cells of the suspensor, DNA contents formed a regular series 256C, 512C, 1024C, 2048C, 4096C and 8192C. By examining the largest suspensor cells at different times in development it was shown that there was a continuing progression toward the highest DNA content, indicating continued synthesis of DNA through development.
