Abstract

Objective To investigate the feasibility of PLGA-poloxamer nanoparticle used as siR-NA carrier.Methods Nanoparticle was prepared by solvent diffusion technique.The diameter,zeta poten-tial and superficial appearance of nanoparticles were observed.Entrapment efficiency of plasmid DNA, in-tegrity of plasmid DNA released from nanoparticles, DNA protection effect, and transfection efficiency were examined.Results PLGA-poloxamer was a globular particle with diameter about (210.1±24.3)nm, ze-ta potential about (-0.43±1.43) mV,and its entrapment efficiency of plasmid DNA was about 31%.Plasmid DNA released from the nanoparticles was almost kept intact.This nanoparticle could protect plas-mid against DNase I digestion and the transfection efficiency was 28%.Conclusion PLGA-poloxamer nanoparticle should be considered as an alternation of siRNA carrier.
