Abstract

The protonation equilibria and conformational changes of a mixture of tyrosine and valine tRNAs were studied by means of spectroscopically monitored acid-base titrations (UV spectrophotometry, circular dichroism and molecular fluorescence) in a working aqueous medium 0.15 M in NaC1 and at 37 °C. Spectroscopic data were analyzed by an Alternating Least Squares (ALS) based multivariate curve resolution procedure which consists of using several chemometrical techniques based on factor analysis (principal component analysis, evolving factor analysis and ALS optimization) [1, 2]. This procedure allows by itself, and without a prior knowledge of the system, the determination of the number of different species present in the system and the estimation of their concentration profiles (quantitative information) and their pure UV, circular dichroism or fluorescence spectra (qualitative information).
