Abstract

Preimplantation genetic testing for aneuploidies (PGT-A) has been introduced into clinical practice to improve outcomes. However, diagnostic accuracy remains controversial, particularly regarding chromosomal mosaicism and de novo segmental variants. Ultimately, the interpretation of these diagnoses is exceedingly complex due to the absence of a precise clinical phenotype. Here, we use an extended embryo in vitro culture model to establish the positive predictive value (PPV) of PGT-A. Our platform allows the culture of human embryos up to 12 days post fertilization (D), providing a unique opportunity for assessing chromosomal (in)stability during human peri-implantation development.
