Abstract
Objective To investigate the expression of microRNA (miR)-129 in prostate cancer and its effect on proliferation of PC-3 cells.Methods The expression of miR-129 in prostate cancerous and corresponding paracancerous tissues of 37 cases of prostate cancer was detected by real-time quantitative polymerase chain reaction (Real-time PCR).Has-miR-129 mimic was transfected into PC-3 cells,then the expression of miR-129 in PC-3 cells 48 h after transfection was detected by Real-time PCR,and proliferations of PC-3 cells after subsequent cell culture for 0,24,48,and 72 h was detected respectively by cell counting kit-8 (CCK-8) assay.Results The expression of miR-129 in prostate cancer tissues (1.088 1 ± 0.578 5) was lower than that in corresponding paracancerous tissues (1.498 6 ± 0.782 8,P < 0.05).The expression level of miR-129 in PC-3 cells after transfection by has-miR-129 mimic was 435 times higher than in control groups (P <0.01).The absorbance value of transfection cells (0.714 7-±0.072 1) after 48 h cell culture was significantly lower than that of control cells (0.878 4 ± 0.057 3,P < 0.05).Conclusion There was low expression of miR-129 in prostate cancer tissues.Overexpression of miR-129 could inhibit proliferation of prostate cancer cells.
