Abstract

Objective To study the expression of bone morphogenetie protein 4 in Adriamyeininduced inhibition in U251 ceils. Methods Application of MTT was used to detect the activity of U251 cell growth in control group and adriamycin group. The formula ( 1-adriamyein group A/control group A) × 100% was used to Calculate U251 cells growth inhibition rate of adriamycin group. Fluorescence quantitative PCR and Western blot were used to detect the expression of BMP4 and Smad4. Results Growth inhibition rates of U251 ceils in Adriamycin group were 10% ,21% ,56% ,49% ,43% at the 24,36,48,60, 72 h. Growth inhibition rate was highest at the 24 h (P 〈 0.05 ). Fluorescence quantitative PCR showed that the expression of BMP4 in Adriamycin group compared with the control group was increased by ( 13.0 ± 0.7 ) % ,the expression of Smad4 increased by ( 35.0 ± 1.0) %. Western blot showed that the expression of BMP4 and Smad4 in Adriamycin group increased by 23% and 38% respectively compared with the control group. Conclusion Inhibition that adriamycin induced in U251 cells, may be implemented by increasing expression of BMP4 and Smad4. BMP4 may be a new target in the treatment of glioma.
