Abstract
Objective To study the expression and significance of microRNA(miRNA,miR)-146a in osteosarcoma. Methods The expression levels of miRNA-146a in osteosarcoma tissue and normal tissue adjacent to carcinoma were detected by real-time quantitative polymerase chain reaction(Real-time PCR). The correlation between survival and miRNA-146a level was analyzed by Cox's proportional hazards regression model.The cell viabiliyt,apoptosis and the levels of Yin-Yang 1(YY1)and Fas were measured after miRNA-146a was transfected into osteosarcoma TE85 cells. Results The expression levels of miRNA-146a in osteosarcoma tissue and normal tissue adjacent to carcinoma were respectively 1. 213±0. 525 and 0. 734± 0. 263.There was a positive correlation between survival and miRNA-146a expression level(P<0. 05).After transfected with 50,100 nmol/L miRNA-146a for 24 h, the cytoactive were respectively(65. 161±5. 528)% and(40. 385±8. 442)%(P< 0. 05) and the apoptosis rates were respectively(38. 671±12. 415)% and (59. 513±9. 058)%(P<0. 05).The YY1 expression levels were significantly decreased(0. 472±0. 014 and 0. 272±0. 005, P<0. 05),and the Fas expression level were significantly increased(0. 292±0. 010 and 0. 584 ±0. 006, P<0. 05).Compared to control group, the changes of cytoactive,apoptosis rates, YY and Fas expression levels after transfected with 50 and 100 nmol/L miRNA-46a for 48 h were similar to those changes in osteosarcoma cells transfected with miRNA-146a for 24 h. Conclusion miRNA-146a may inhibit the proliferation of osteosarcoma cells,and simultaneously promote the apoptosis of osteosarcoma cells, probably by down-regulating the YY1 expression and up-regulating the Fas expression.
