Abstract

Objective Using quantum-dot-fluorescence labeling for detecting the characteristic of prostate stem cell antigen(PSCA)protein expressed in the transitional cell carcinoma(TCC)of bladder.Methods Fifty-two cases of TCC contained 34 superficial TCC(STCC,stages Ta-T1)and 18 muscle-invasive TCCs(ITCC,stages T2-T4).Quantum-dot-fluorescence labeling analysis of PSCA protein expression was performed on the entire specimens and each of the cases set up a negative control group.The expression of PSCA protein Was observed at 4 time points within 1 month,and the level w8s scored in 4 grades by assessing the intensity of fluorescence.Results The surface of tumor cells express orange red fluorescence under the stimulating of blue light,the cytoplasm and nucleus with no expression.With obviously control,the negative controls express spontaneous green fluorescence.The expression of fluoreseence in STCC Was more uniform than that seen in ITCC,with fluorescence detected 97.1%(33/34);however.the ITCC also detected 77.8%(14/1 8).Higher level of fluorescence expression correlated with the increasing tumor grade.The strong fluorescence expression in grade Ⅱ was 43.3%and 59.1%in grade Ⅲ.The level of the fluorescence change Was not significant in 2 weeks,in comparing.the fluorescence hardly keep stabilization last for 1 month,there Was 4 specimens with the fluorescence drop to 0 and none 3+ level specimen exist.However,the last specimens distributed in the+and 2+by fluoresceBee level could still be seen.Conclusion PSCA is a valuable tumor marker,particularly superficial with higher grade tumor,which prompts that it may be a useful target for early diagnosis and monitor recurrence of TCC of bladder.
