Abstract

Objective To investigate glypican-5 (GPC-5) expression in lung squamous cell carcinomas and its clinical significance.Methods GPC-5 was detected in 64 cases of lung squamous cell carcinoma and corresponding normal lung tissue by real-time quantitative polymerase chain reaction (PCR),which was also validated in human lung squamous cell carcinoma tissue microarrays (TMA,containing 75 cases of squamous cell carcinoma tissue and adjacent tissue) by using immunohistochemistry.Results The expression levels of GPC-5 mRNA and protein in squamous cell carcinomas were significantly lower than in matched normal lung tissues (P < 0.001).The expression levels of GPC-5 mRNA and protein in the lymph node metastasis group were lower than those in the non-metastasis group (29.0074 ±6.4013 vs 11.4849 ±2.5066 folder,P <0.05).The expression levels of GPC-5 mRNA in the pTNM Ⅱ A-B group were lower than those in the pTNM Ⅰ A-B group (28.0566 ± 3.8333 vs 11.9384 ± 6.6651 folder,P < 0.05).Spearman analysis revealed that the expression levels of GPC-5 mRNA were significantly correlated with lymph node metastasis (r =0.270,P < 0.05).Conclusion GPC-5 may be a new tumor suppressor gene,and is closely related with the lymph node metastasis of lung squamous cell carcinoma.
