Abstract
Objective To evaluate the long intervening noncoding RNA (LincRNA)-HOX transcript antisense RNA (HOTAIR) expression in colorectal cancer and its biology function of proliferation and metastasis in human colorectal cancer cell line LoVo. Methods Real-time quantitative polymerase chain reaction (Real-time PCR) was applied to analysis the expression of HOTARI in paired colorectal cancer tissues and para-cancer normal colorectal tissues. And the HOTAIR was knocked down by small interfering RNA (siRNA) in LoVo cells. Methyl thiazol tetrazolium (MTT), wound healing and transwell assay were applied to evaluated the cell proliferation and invasion ability. Results HOTAIR expression were 7.96±4.15 and 4.47±2.70 for cancer tissue and para-cancer normal colorectal tissue respectively with statistical differenc (t=4.520, P 0.05). Conclusion HOTAIR was high expressed in human colorectal cancer tissue and down-regulation of HOTAIR in colorectal cancer can promote cell migration/invasion.
