Abstract

Objective To study the mechanism of apoptosis induced by As2O3 in laryngeal squamous carcinoma Hep-2 cell strain.Methods The growth inhibition of chemotherapeutical drugs-As2O3 on Hep-2 cells was assayed with the growth curves and MTT test.TUNEL stain and electron microscopy were used to detect the apoptosis of Hep-2.Expressions of Survivin gene were compared between As2O3-induced Hep-2 cells and the control cells by using Western-blot.Results As2O3 obviously inhibited the growth of laryngeal squamous carcinoma Hep-2 with dose and time-dependent.Apoptosis was a major way of Hep-2 cell death induced by As2O3.The decreased expression of Survivin gene after As2O3 treatment was also confirmed by Western-blot.Conclution As2O3 could inhibit the growth and ihduce the apoptosis of laryngeal squamous carcinoma Hep-2 cell strain by restraining the expression of Survivin.
