Abstract

Context and objective: Bauhinia acuminata Linn.is used in inflammatory diseases by the local traditional healers of Purulia district of West Bengal, India. In the present study, an attempt has been made to dissect out the probable mechanism of action of the anti-inflammatory profile of Bauhinia acuminata stem bark by using LC-MS/MS along with various in vivo experimental models. Material and method: The hydro-alcoholic extract of the stem-bark of Bauhinia acuminata Linn.(HEBA) was subjected to preliminary phytochemical analysis and evaluated for acute toxicity study, carrageenan induced rat paw edema (analysis of PGE2 was done by LC-MS/MS) and acetic acid-induced peritoneal inflammation. Evaluation of analgesic activity was performed in models like radiant heat induced algesia (tail flick method) and acetic acid induced writhing. Results and conclusions: The extract showed significant inhibition in increase in paw volume of the right hind paw as compared to control group in carrageenan-induced rat paw edema. In acetic acid-induced peritoneal inflammation, pre-treatment with the extract demonstrated significant inhibition of acetic acid induced peritoneal protein exudation and also reduction in peritoneal leukocytes count. In the tail flick test, HEBA hardly produced any significant effect but it has shown significant inhibition against acetic acid induced writhing in mice. The effect might be attributed to the combined effect of phytoconstituents like flavonoids and glycosides and others present in the extract. In conclusion, an optimized, highly sensitive and specific LC-MS/MS technique for the detection of main arachidonic acid metabolites formed in the rat paw tissue in the late phase of the carrageenan induced inflammation model is presented here along with the other in-vivo models. Overall, these results undoubtedly confirm the anti-inflammatory activity of the hydro-alcoholic extract of the stem bark of the Bauhinia acuminata Linn., along with an indication of its mechanism of action.
