Abstract

Switching code at Tyr66. We report the expansion of the genetic code in a mammalian cell line by using various four-base codons. These were derived either from an amber stop codon or rare codons from mammalian cells. Among the four-base codon/anticodon pairs examined, the UAGG/ccua pair was the most efficiently decoded and expressed the highest amount of enhanced green fluorescent protein (EGFP; see scheme).
