Abstract

The antibiotic penicillin is being a major pharmaceutical compound after several years of use. The present research work focuses on the selection of suitable low cost substrate from rice based residues like grains, husk and bran for the seed culturing of Penicillium chrysogenum by solid state fermentation (SSF) and optimization of medium for hyper production of penicillin V (Pen V) by P. chrysogenum under submerged fermentation (SmF). In SSF, significantly highest number of total spores and viable spores were augmented in rice bran media than the rest. The effect of different carbon, nitrogen sources, pH, temperature, incubation time and various concentrations of precursor amino acid (l-valine) were studied for hyper production of Pen V by P. chrysogenum. The results revealed that in SmF, the enriched rice bran medium has had increased the production yield of Pen V at a highest level of 22.1 g/L than the synthetic production medium 19.8 g/L. Hence, the Pen V produced from the enriched low cost rice based substrate is being an alternate for next generation production.
