Abstract

Cell-free extracts containing nuclei and chloroplasts were prepared from turnip yellow mosaic virus-infected Chinese cabbage leaves from plants previously treated with 32P-orthophosphate. After fractionation by sedimentation into discontinuous sucrose density gradients, the 32P-labeled double-stranded viral RNA in the extracts distributed as did the chlorophyll-containing chloroplast fractions rather than the DNA-containing nuclei. The results support our previous suggestion that double-stranded turnip yellow mosaic virus RNA is associated with the chloroplast fraction from virus-infected Chinese cabbage leaf, and not with nuclei.
