Abstract

1 Indoramin has been reported to reduce blood pressure, antagonise experimental cardiac dysrythmias, and increase peripheral blood flow in patients and normal volunteers. 2 In this study the effect of the drug on heart rate and rhythm, central venous and arterial blood pressures, cardiac output, and leg blood flow was determined in 11 healthy volunteers. 3 Indoramin (5–20 mg) given intravenously resulted in increases in leg blood flow amounting to 260% in the foot and 40% in the calf as estimated by strain gauge plethysmography. No significant change in heart rate or cardiac output was noted, but central venous pressure was diminished (2mm Hg) and arterial pressure decreased (systolic 10mm Hg and diastolic by 6 mm Hg). 4 Haemodynamic responses to isoprenaline (1–4μg) were not influenced by indoramin but those to noradrenaline (6–20μg) were diminished suggesting that these responses were due to α-adrenoceptor blockade.
