Abstract
Objective To evaluate the antioxidant effects of compound Chinese medicine Tangbikang's on diabetic peripheral neuropathy rat. Methods The rats of diabetic peripheral neuropathy (DPN) were induced by streptozotocin intraperitoneally. The DPN rats were divided into 5 groups:model group, diabetic peripheral neuropathy rats treated with mecobalamin,DPN rats treated with the low dose of Tangbikang,DPN rats treated with the medium dose of Tangbi-Kang,and DPN rats treated with the high dose of Tangbikang.MDA and SOD levels in plasma, liver, muscels and nervous tissues were detected after continuously for 12 weeks, and the ultrastructure of sciatic never in rats were observed. Results The different dose of Tangbikang could reduce the contents of MDA in liver [(8.47± 1.82,7.57± 1.45,8.71 ± 1.51)nmol/ml] compared with model group [(10.42±2.05) nmol/ml] with significant difference (P<0.01,P<0.05),increase the contents of SOD in live r[(21.12± 1.52,22.29±2.21,19.29± 1.10) U/ml],compared with model group[(18.24± 1.98) U/ml] with significant difference (P<0.01,P<0.05),reduce the contents of MDA in plasma, muscels and nervous tissues, compared with model group, with significant difference (P<0.05),and increase the contents of SOD in plasma, museels and nervous tissues on DPN rats, compared with model group, with significant difference (P<0.05).After treated with Tangbikang, the results of transmission electron microscopy showed that the widely myelin sheath and loose structure of myelinated nerve fibers of sciatic nerve were improved significantly, plaque demyelinating and segmental demyelination phenomenon were reduced significantly, the vacuolus defects was rare, neurofilaments, neurotubule was visible clearly, the structure of myelin sheath tended to be complete and regular. Conclusion Tangbikang has antioxidant effect. It can reduce the damage of peripheral nerve structure and function by oxidative stress, and protect nerve of DPN rats. Key words: Tangbikang; Diabetic peripheral neuropathy; Rat; Oxidative stress
