Abstract
Objective To investigate the effects of different concentrations of thrombin,trasylol on the release rate of the system in vitro by establishing a stable fibrin gel(FG) release system, simultaneously discuss the possibility of FG as growth factors delivery system. Methods Different concentrations of thrombin,trasylol FG release system was established in vitro with fibrinogen, thrombin, trasylol, calcium chloride. The influence of growth factors on this FG release system was studied, four growth factors(platelet-derived growth factor-AB,vascular endothelial growth factor, epidermal growth factor, transforming growth factor-β1) were added in this system as the experimental group, non growth factors as the control group, and the dissolution phenomenon of these two groups in 37℃water bath was observed. The stabilities of biological activities of these growth factors in the release system were studied by measurements of the concentrations of growth factors in the experimental group by ELISA assay. Results When thrombin was at the same concentration, the higher the concentration of trasylol was, the slower dissolution rate of the FG release system was(P 0.05). Furthermore, there was no significant difference in the concentration of growth factors daily dissolution(P> 0.05). Conclusions The FG release system can dissolve in a steady, slow and well-distributed way. Under the condition of low concentrations of trasylol, the increase of concentration of thrombin can accelerate the dissolution rate of FG release system. In the different way, at high concentrations of thrombin, the increase of concentration of trasylol can delay the dissolution rate of this system. In addition, FG will be not affected by growth factors, and can maintain its stable biological activity, which indicates FG can be used as growth factors delivery system.
