Abstract

Objective To analyze the differential protein expression by high resolution and reproducibility two -dimensional electrophoresis profiles in the invasion and noninvasion pituitary adenomas. Methods The proteome from the invasion and noninvasion pituitary adenomas tissues were separated and identified by using a series of methods,including immobilized pH gradient two - dimensional polyaerylamide gel electrophoresis, silver staining, ImageMaster 2 DE analysis software, peptide mass fingerprint based on matrix -assisted laser desorption/ionization time of flight mass spectrometry ( MALDI - TOF - MS) and database searching. Results The good 2 DE pattern of the invasion and noninvasion pituitary adenomas tissue including resolution and reproducibility were obtained. The map of the invasion pituitary adenomas tissues was compared with the noninvasion pituitary adenomas. Thirty proteins were analyzed, some of which have the relationship of oncogenes, and others were the regulation proteins, which adjust the cell cycle and signal transduction. Conclusion These data suggest that we constructed a high resolution and reproducibility of two - dimensional electrophoresis profiles from the invasion and noninvasion pituitary adenomas and analyze the differential protein spots, which will be taken advantage to establishing the differential proteomic expression profiles. These data will be used for establishing the invasion pituitary adenomas proteome database.
