Abstract
Current ACMG guidelines recommend periodic reassessment of variant classifications with current information. However, with the ever-increasing size of a laboratory's variant database, periodic reassessment can become a burden. Variants of uncertain significance (VUS) are the classification category most likely to change on reassessment due to increased population data, new studies regarding pathogenicity, and evolving classifications standards. ClinVar is a government website developed by the National Center for Biotechnology Information that allows laboratories to submit variant classifications.
