Abstract

The large liposome with about 5-50µm in diameter,which was composed of Span80(sorbitan monooleate) as a main component and contained a little amount of phospholipid, could be prepared by the application of the second-step emulsification for providing a W/O/W emulsion.4) The characteristics of the lipid-membrane of the large liposome under various preparation conditions were examined. Moreover, the applicability of the liposome for encapsulated enzyme was examined.The model reaction in the liposome was NAD+-recycling catalyzed by malate dehydrogenase (MDH) and yeast alcohol dehydrogenase (ADH). ESR measurements were also carried out to clarify where the enzyme protein exists in the liposome:in the inner aqueous phase or in the lipid-membrane.
