The DNA topological challenges generated by cellular manipulation of extremely long DNA fibers remain poorly understood. In this issue of Molecular Cell, Hildebrand etal.1 describe how mitotic chromosomes are self entangled and that disentanglement requires TOP2 activity in late mitosis.
