Abstract
Suppression of a conditioned avoidance response in rats by 2 mg/kg chlorpromazine HCl (CPZ) was enhanced (230%) after treatment with 50 mg/kg acetazolamide. This enhanced pharmacological response in the acetazolamide-treated rats was accompanied by increased brain levels of 35S after administration of chlorpromazine. In parallel experiments, an analogue of chlorpromazine, CL-13,850. which possesses no carbonic anhydrase inhibitory activity, had no influence on the pharmacological effect or the brain levels of 35S after administration of chlorpromazine. The data suggest the enhanced pharmacological response following acetazolamide treatment is the result of increased brain levels of chlorpromazine brought about by carbonic anhydrase inhibition.
