Abstract

In this study we compared the efficiency of direct colony PCR of bacterial and fungal DNA with Taq DNA polymerase and MightyAmp DNA Polymerase. The results show that whilst only 44.0 % of bacterial species representing 13 genera and 11.1 % of fungal species representing 18 genera could be amplified by DCPCR with Taq DNA polymerase, all 25 bacterial species and 27 fungal species including 1 biotrophic fungus Puccinia cannacearum could be amplified by DCPCR with MightyAmp DNA polymerase.
