Abstract

Analysis of rare transcripts in tissues or comparative gene expression analysis is one of the main goals of researchers worldwide. Since the development of real time polymerase chain reaction (PCR) for such purpose, there has been raising interest to apply this tool in molecular biology systems, including basic veterinary. We developed an efficient and cost effective real time assay, particularly for rat’s P-glycoprotein (P-GP) gene, which play an important role in complicated drug transporter systems. We could perform a reliable assay with high efficiency (>100) and low target (>40 ng total RNA) for the P-GP gene. The home-made primer design is characterized by its high target-specificity. Further, we compared two popular real time methods, cyber green and Taq man, and found that both systems deliver suitable results; but the taq man system seems to be more effective in the case of low or limited target (RNA concentration).   Key words: P-Glycoprotein (P-GP) gene, rat, real time polymerase chain reaction (PCR), expression analysis, low target.
