Abstract

The aim of this study is to predict possible apoptotic pathway and elucidate the multi-targets’ pharmacological mechanism of Chinese medicine in preventing and treating cardiovascular diseases through apoptosis pathway. Literature mining methods is applied to analyze literatures related to Chinese medicine with cardiomyocyte apoptosis in Chinese National Knowledge Infrastructure (CNKI) and pubmed database from the year 2005 to 2012. Drug targets cluster analyses were conducted and targets related networks were established by combining Herbal Ingredients' Targets database (HIT) with GenCLiP 2.0 software (Human Gene functions and network analysis). Meanwhile, this study provides a new method to predict potential targets of Chinese medicine in treating cardiovascular diseases and identify the active continent.   Key words: Cardiomyocyte apoptosis, traditional Chinese medicine, gene cluster analysis, network analysis.

Highlights

  • Cardiomyocyte apoptosis has been identified as an essential process in the progression of cardiovascular diseases (Vanessa et al, 2005; Kazuhiko et al, 2008; ZhiQing and Jacob, 2002; Peter et al, 2000)

  • By applying GenCLiP 2.0 online analysis software (Human Gene function and network analysis is an online analysis database which retrieves literatures, does cluster analysis and constructs network based on different keywords) to do apoptotic cluster analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway cluster analysis with gene symbols retrieved from Herbal Ingredients' Targets database (HIT) database, we tried to find specific apoptotic pathways and the multitargets network of those apoptosis related Chinese medicines

  • Thirty-six (36) Chinese medicines and 44 formulas were found related to cardiomyocyte apoptosis

Summary

Introduction

Cardiomyocyte apoptosis has been identified as an essential process in the progression of cardiovascular diseases (Vanessa et al, 2005; Kazuhiko et al, 2008; ZhiQing and Jacob, 2002; Peter et al, 2000). Apoptosis cells are maximum in the edge area of myocardium, whereas in control myocardium, only rare cardiomyocytes stained positive (Rabkin, 2009). In traditional Chinese medicine (TCM), the etiology and pathogenesis generalize cardiovascular disease as “heart obstruction”, “cardiac edema”, “asthma”, “palpitation” and so on. TCM has definite clinical therapeutic effect on trea-ting cardiovascular disease, especially on improving the life quality of patients. Through literature mining we found that researches on Chinese medicine pharma-cological mechanism of apoptosis are lack of pertinence. Though many researches on mechanism of Chinese medicine have been conducted on apoptosis, they merely focused on the important molecules such as caspase-3 and Medicine Chinese herb
