Abstract

Summary Objective : to evaluate the possible influence of different concentrations of asparaginase (ASP), dexamethasone (Dex) or the combination ASP/Dex on porcine endothelial cells we measured von Willebrand factor (vWf), tissue-type plasminogen activator (t-PA), plasminogen activator inhibitor 1 (PAI-1) and thrombomodulin (TM) in culture supernatant fractions. Results : vWf significantly increased in cells treated with 100 IU/ml ASP, but showed no difference in cells incubated with 1000 or 500 IU/ml ASP. t-PA concentrations were significantly increased in cells treated with 1000 and 100 IU/ml ASP. Dex alone and ASP/Dex resulted in a glucocorticoid dose-dependent increase of vWf and t-PA. PAI-1 and TM could not be determined in this cell line. Conclusion : these data provide evidence, that endothelial damage, occurring during treatment with ASP combined with Dex is mainly caused by glucocorticoid administration.
