Abstract

BackgroundEarly diagnosis of cancer can help not only in saving precious lives but also in reduction of the disease management expenses due to the effective and shorter anti-cancer treatment. Heat shock protein 70 (HSP70) is a cancer biomarker whose level is increased in a plethora of cancerous tumors. Therefore, this work is focused on the development of cost-effective label-free biosensing over a novel plastic chip electrode (PCE) platform for highly sensitive and selective detection of the HSP70 biomarker. MethodsThe gold-coated surface of PCE was modified with 4-aminothiophenol and glutaraldehyde linking chemistry, and further immobilized with a bioreceptor anti-HSP70. Further, sensing experiments were performed using differential pulse voltammetry. FindingsThe results confirmed the successful surface modifications and immobilization of the anti-HSP70 antibody on the PCE platform. PCE electrochemical biosensor displayed the highly sensitive detection of the HSP70 in the wide linear range of 0.01-1000 ng/mL and a low limit of detection (LOD) of 3.5 pg/mL at 3σ. Also, HSP70 detection from the serum samples with excellent sensitivity, accuracy and specificity was demonstrated. Overall, the novel PCE biosensor has demonstrated great promise for highly sensitive and selective detection of the HSP70 cancer biomarker.
