Abstract

THERE have been a number of studies on the effects of ultra-violet microbeam irradiation of delimited areas within cells in culture. In experiments with mitotic cells1–10, it has been found that irradiation of a chromosome produces a loss of DNA and histone at the site of irradiation3,4,9, and also that the spindle diminishes in size or disappears temporarily following the irradiation of any part of the cell1,2,5–8,10. In experiments with interphase cells11,12, some authors have reported that about two-thirds of the labelling of cytoplasmic RNA is suppressed by nucleolar irradiation11, while extranucleolar nuclear irradiation results in a progressive inhibition of DNA synthesis for several hours after the irradiation12. We have carried out an autoradiographic study on the effects of ultra-violet microbeam irradiation of different cellular sites on synthesis of RNA, DNA and protein in order to compare the pattern of inhibition in a single cell type.
