Abstract

Objective To investigate the effects of silencing lysyl oxidase-like 2 protein (LOXL2)gene on pancreatic cancer cells.Methods LOXL2-short hairpin RNA (shRNA) plasmid was constructed by using lipofectamine,and transfected into pancreatic cancer Panc-1 cells.The changes of LOXL2 protein expression in pancreatic cancer panc-1 cells before and after transfection with LOXL2-shRNA were assayed by using Western blotting.Transwell invasion assay was used to assess the in vitro invasive ability of pancreatic cancer cells.Adhesive capacity of pancreatic cancer cells was measured by methyl thiazol tetrazolium (MTT) assay.Results The grey-scale value of LOXL2 protein in Panc-1 cells in LOXL2-shRNA transfection group was (0.32 ± 0.01),the number of migrating cells was (32.2 ± 3.8) and adhesive capacity was (75.4 ± 3.2)%,which were significantly reduced as compared with those before transfection [1.05 ± O.05,68.6 ± 3.5,and (90.3 ± 2.1) %,P < 0.05].Conclusion Silencing LOXL2 gene can inhibit the invasive ability and adhensive capacity of pancreatic cancer Panc-1 cells.
