Abstract

Objective To investigate the effect of the mannitol-glycerol injection (containing 15% mannitol and 15% glycerol) on hemolysis of red cells in rabbits and rats.Methods By the test tube method in vitro to observe hemolytic effect of mannitol-glycerol injection in the normal rabbit red blood ceils.The neighborhood-a oxidation benzidine chromogenic method was used to determine the levels of plasma free hemoglobin content.The SD rats'red blood cell hemolysis in vivo was determined.The plasma free hemoglobin and hemobilirubin was determined.Results Compared with the sodium chloride group,the 1.6 ml,2.0 ml tubes'OD values of RBC + 5 % glycerol group by testing in spectrophotometric method,and the 1.6 ml,2.0 ml tubes'OD values of 5% glycerol group by testing in ortho-tolidine method had statistically significant differences (P <0.01).There were significant differences of each tube in 5% glycerol + RBC group (P<0.01).While RBC + mannitol glycerol group and mannitol,glycerol + RBC group compared with the Sodium Chloride group showed no significant difference (P >0.05).By 6,10 ml/min dose rate,30 ml/kg volume intravenously administered to rats corresponding substances,there was no statistical significance between mannitol and glycerin injection group's plasma free hemoglobin and hemobilirubin with sodium chloride group after 2,24 hours (P >0.05); 5% glycerol group's 24h hemobilirubin compared with the sodium chloride group has statistically significant differences (P < 0.05).Conclusions The mannitol-glycerol injection had no hemolytic effect on erythrocytes in vivo and in vitro.
