Abstract

The goal of our experiments was to prove whether the monomer IH 764-3 (a purified preparation from Chinese herb of Huoxue Huayu) showed the release activity of tissue plasminogen activator or not. 1. The rat hind legs were perfused with IH 764-3 for 5 min. The PA activity in the perfused fluid was estimated by fibrin plate assay. It was found that the PA activity was increased with dose dependent from 35 to 280 μg/ml, showed the obviously increased activity at 70 μg/ml and 140μg/ml. For determination of the PA type, the collected perfused samples were analyzed by SDS-PAGE and fibrin-autography technique. It showed a lysis zone at 67KD. When the McAb against t-PAl was added into the indicator gel and then there was no lysis area at 67KD zone. 2. IH 764-3 was incubated with the confluent primary endothelial cells from bovine aorta under serum-free medium for 4 hrs, The t-PA activity was measured with the chromogenic substrate (S-2251) The results showed the t-PA activity was obviously increased and with dose-dependent.
