Abstract
Objective To investigate the effects of hyperbaric oxygen (HBO) on rats with premature ovary failure (POF) induced by triptergium wilfordii polyglycisidium (TWP).Methods Fifty female SPF rats were randomly divided into 5 groups:the normal group,the model group,the control group,the hormone replacement therapy (HRT) group and the HBO group,each consisiting of 10 animals.Except the normal group,all the other groups were given orally triptegium wilfordii polyglycisidium (TWP) to establish premature ovary failure model.Fourteen days after intragastric administration of TWP,serum samples from the normal and model groups were taken,then the animals were sacrificed and uterus and ovary were collected for detection.The animals in other groups were treated differently,and 40 days after establishment of the model,serum samples were taken,then the animals were also sacrificed and uterus and ovary were collected for the detection of related parameters.Serum estradiol (E2) level,changes in estrous cycle,uterine and ovarian weight,endometrial thickness and ovarian size were observed and analyzed.Results After intragastric administration of the drug,estrous cycle of rats was disturbed in various forms.Following treatment with HRT and HBO,estrous cycle all gradually recovered to a more or less extent,while estrous cycle of the rats in the control group still remained in a state of disorder.Serum E2 level,endometrial thickness and organ weight index in the model group were significantly lower than those of the normal group (P < 0.05).Besides,follicular number in ovary unit area was significantly less than that of the normal group,while atretic follicular number was obviously more than that of the normal group (P < 0.05).Ovary area in the model group was smaller than that of the normal group (P > 0.05).Forty days after intervention,serum E2 level in the HBO group was higher than that of the control group and the HRT group (P < 0.05).Endometrial thickness and uterus weight indexes in the HBO group and the HRT group were significantly higher,as compared with those of the control group (P < 0.05),and the levels of the HBO group were even higher than those of the HRT group (P < 0.05).Ovary unit area and follicular number of the HRT group were either larger or greater than those of the HRT group and the control group,however,statistical significance could only be noted,when they compared with those of the control group (P < 0.05).Atretic follicular number of the HBO group and the HRT group was significantly less than that of the control group (P < 0.05).Conclusions It was feasible to establish the rat model of POF by intragastric administration of TWP.HBO could improve the function of ovary in rats with premature ovary failure,elevate serum E2 level,increase uterus and ovary weight and endometrial thickness as well.
