Abstract

The effects of different types of bioreactor and NAA concentrations on growth and tropane alkaloid production in adventitious root cultures of Atropa belladonna were investigated. The growth of adventitious roots was markedly stimulated and callus induction was inhibited in an aeration-agitation bioreactor in comparison to an air-sparged biorector. Although tropane alkaloid content in dry cell mass was decreased in roots cultured in the aeration-agitation bioreactor, their productivity was stimulated because of the high biomass.The most efficient conditions for tropane alkaloid production included the use of an aeration-agitation bioreactor containing modified MS medium supplemented with 0.3 mg/l NAA and 60 g/l sucrose. The results of this experiment suggested the usefulness of aeration-agitation bioreactors for scaling-up of various root cultures.
