Abstract

Objective To investigate the effects of recombinant adeno-associated virus (rAAV)-CD151 on the expression of the integrin α3β1/α6β1 in human umbilical vein endothelial cells (HUVECs). Methods rAAV-CD151, rAAV-CD151-QRD and rAAV-CD151-ARSA were produced and transfected into HUVECs directly. Protein levels of CD151 were detected. And the protein expression of integrin α3,α6 and β1 on the surface of HUVECs were measured by western blot and flow eytometry. Results Transfections of rAAV-CD151 increased the expression of CD151( P<0.01 )and integrin α3β1/α6β1 ( P <0.05). On the contrary, transfectians of rAAV-CD151-QRDand rAAV-CD151-ARSA only increased expression of CD151 ( P<0.05 ), but didnt increase expression of integrin α3β1/α6β1. Conclusion CD151 gene transfection could promote the expression of integrin α3β1/α6β1. The mechanism of regulating integrin α3β1/α6β1 may lie in allosterism of an extracellullar CDI51 site (QRD194-196) and C -terminal Ves. transportation target motif(YRSL245-248).
