Abstract
Tris-based diluent is superior to citrate-based diluent for bull semen dilution. Caffeine (1,3,7-trimethyl-2,6-dioxy-purine), a cyclic nucleotide phosphodiestrase inhibitor, maintained good semen criteria when added to the semen at levels of 10 and 20 m M; however, higher levels caused a deleterious effect. This was emphasized by assessments of sperm penetration into cervical mucus. Incubation of the semen at 37°C for 6 h caused gradual decreases in sperm motility, fructose content and pH.
