Abstract

1. 1. Bunamidine caused a dose-dependent decrease in spontaneous heart rate, coronary flow and isometric systolic tension, but increased diastolic tension at 10,000 ng/ml. 2. 2. Bunamidine altered electrical activity by prolonging PR, QRS and QT intervals. It also produced partial heart block and ventricular standstill. 3. 3. Bunamidine diminished cardiac glycogen, ATP and creatine phosphate and increased tissue content of d-glucose-6-phosphate, l-(−)-glycerol-1-phosphate and lactate. Conversion of phosphorylase b to a was increased. 4. 4. The increase in phosphorylase a activity and isometric systolic tension caused by epinephrine (0.5 μg) was the same in control and bunamidine-perfused hearts.
