Abstract

Objective To observe the expression of P-selectin in lung tissue following limb ischemia reperfusion in rats and investigate the role of endothelin-A receptor antagonist BQ123 in it.Methods Male Wistar rats were randomly divided into three groups (n =10):control group,LIR group, and BQ123 treated group.ELISA method was used to detect the concentration of P-selectin.PaO2,the total protein content in bronchoalveolar lavage fluid, the levels of myeloperoxide and endothelin-1 in lung tissue were measured. The pathological changes of lung tissue were observed. Results Compared with LIR group, the level of P-selection in lung tissue was decreased,PaO2 was increased, the protein content in bronchoalveolar lavage fluid, the contents of myeloperoxide and endothelin-1 in lung tissue were lower in BQ123 group (all P <0.01).Histopathological examination showed that the degree of lung injury was alleviated in BQ123 group. Conclusions BQ123 inhibits the formation of P-selection to have protective effects on lung tissue after limb ischemia reperfusion in rats. Key words: Reperfusion injury; P-selectin; Endothelin-1
