Abstract
To explore the influence of Asparagus polysaccharide on Band 3 proteins and glycophorin A(GPA) of S180 mice erythrocyte membrane by contrasting the groups treated with deferent ways,The experiment was equipped with 5 groups: normal control group (physiological saline), negative control group (physiological saline), Asparagus polysaccharide group (25mg/kg), Asparagus polysaccharide group (50mg/kg) and Asparagus polysaccharide group(100mg/kg).The content of sialic acid(SA) was determined by spectrophotometric method; the negative charges were detected by high performance capillary electrophoresis(HPCE); the relative content of glycophorin A and Band 3 were separated by sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE; anion transportation function of Band 3 was examined by the fluorescence spectrophotometric method;Flow Cytometer(FCM) was needed to observe the potential of erythrocyte membrane. In the result, Asparagus polysaccharide increased the content of sialic acid; advanced the migration time; improved the relative content of GPA and Band 3; obviously enhanced the CI- transportation function of erythrocyte Band 3; and caused the membrane potential obviously to elevate, Asparagus polysaccharide has the simultaneously influence on the functions of glycophorin A and Band 3 which are closely connected. It is assumed that the anti-tumor function of Asparagus polysaccharide partly concerns function adjustments of glycophorin A and Band 3 from S180 mice erythrocyte membrane.
