Abstract
Summary Aphidicolin markedly reduced progeny virus production in SC-1 cells newly infected with murine leukemia viruses. Once the viral DNA was integrated into the host cell DNA, virus production was not inhibited by the drug. Southern blotting analysis of unintegrated viral DNAs obtained from aphidicolin-treated cells revealed that large closed circular DNA disappeared, although linear and small closed circular DNAs remained. The size of fragments obtained by restriction endonuclease digestion of linear DNA was not altered by the aphidicolin-treatment of the cells. These results indicate that an aphidicolin-sensitive site exists in the process of the formation of large closed circular DNA.
