Abstract
Regulation of cellular differentiation has been suggested to play a major role in the mechanisms of chemical carcinogenesis as well as in the therapeutic effects of anticancer drugs . Therefore , the establishment of differentiation assays appears important in oncology research . Leukemic cell lines have been used as experimental models to study the differentiation-inducing effect of cytostatic compounds in neoplastic cells . Experiments with anthracycline antibiotics in Friend erythroleukemia cells and HL-60 cells showed 1] a structure-specific induction of differentiation and 2] an early differentiation-specific down-regulation of c-myc and c-myb mRNA levels. These results suggest that the induction of differeniation by anthracyclines occurs through specific biochemical mechanisms and that increased cell maturation is not based on a general suppression of cell proliferation. Therefore, the specific induction of differentiation in certain leukemic cells may be a therapeutic advantage of differentiation -inducing anthracyclines such as aclacinomycin A and related compounds.
